I would like to hide coupons, but allow our sales people to be able to add a $25 order level discount at their discretion.

Once I have created a coupon (e.g. 25dollarsoff), can you please show how to apply this to the cart in the querystring?

The SC user guide explains how to add a product to the cart (with a coupon) via a querystring, but I would like know whether a coupon can be applied with a querystring on the Cart page.

It sounded like you were asking to apply the coupon before the Product was added to the Cart anyway, glad you found the solution.
