For a dropdown list variant adjust the sort order so the variant you want as default is the first variant item. For radio button lists and checkbox variants there is no option in the cart to default one radio button or checkbox as selected, however a possible solution when setting up your variants is to check the required checkbox which will require the user to select a variant before adding the item to the cart.
